Background

Sildenafil (Viagra) is a prescription drug for treating erectile dysfunction and its mechanism of action is still under debate. However, the main study of Viagra’s efficacy and safety is by comparing its effect on blood pressure in the subjects with erectile dysfunction (ED) and healthy volunteers. This study was undertaken to compare the effect of Viagra and its immediate-release form on blood pressure (BP) in ED subjects. Methods: This was a double-blind, randomized placebo-controlled trial. Inclusion and exclusion criteria were based on the European Society of Cardiology (ESC) clinical practice guidelines. Patients were randomly assigned to Viagra (n=1,844) or placebo (n=2,064). Blood pressure (BP) was measured before and after treatment at baseline, 3, 6 and 12 hours. Blood pressure (BP) was calculated using the formula: BP [mm Hg] × 100. The study was conducted at the end of the 12-week study, before and after the start of Viagra treatment. Mean ± SD of BP were 4.1±1.9 (mean±standard deviation), 2.9±1.6 (mean±standard deviation) and 3.0±2.6 (mean±standard deviation). Sildenafil was used as an initial drug and was designed to be administered orally in a dosage of 25 mg/day. Following a single dose of Viagra 25 mg, the dose was titrated to a maximum of 100 mg.
